Production process of mother culture of oyster mushroom test tube

1. Recipe of mother culture medium
Take 200 grams of potatoes for juice, 20 grams of glucose powder, about 20 grams of agar, and 1000 ml of water. Potassium dihydrogen phosphate 3 grams, magnesium sulfate 1.5 grams, peptone 3 grams.
 2. Preparation process of mother culture medium
Peel the potatoes and slice them, boil them in 1000ml of boiling water for about 30 minutes, filter them with gauze to get the juice after cooking. (Note that the water must be enough for 1000 ml, if it is not enough, you can make it up with boiling water). Pour the potato juice into the container with the other ingredients and stir as you pour to coat well.
 3. Fill the test tube
Inhale the mixture into the test tube, about one-third of the test tube, and plug the test tube mouth tightly with cotton. Wrap five or six of the packed test tubes with kraft paper and tie them with ropes. Do not cover the lower part for later operation and observation. The bundled test tubes were placed in an autoclave for autoclaving.
 4. Disinfection
The temperature of the pressure cooker is controlled at about 120 degrees, and the disinfection takes about 30 minutes. After taking out the test tube, place it obliquely while it is hot, so that the liquid in the test tube has a slope after solidification, which is conducive to the growth of mycelium.
 5. Inoculation
Take fresh oyster mushrooms, cut off a small piece in the middle with a knife, and put them into test tubes for cultivation. The whole process must be operated in a sterile environment, and the entire instrument must be sterilized with 75% alcohol.
 6. Incubation
Put it in an incubator and keep the temperature at 25 degrees, and the mother species of Pleurotus ostreatus will be successfully cultivated in about ten days.
